| I. Background | |
| 血小板樹突狀細胞抗原 2(BDCA2)是一種僅在人 pDC 表面表達的C型凝集素。BDCA2 由一個糖基識別的胞外域、一個跨膜區(qū)和一個短的不含任何信號轉(zhuǎn)導(dǎo)結(jié)構(gòu)域的胞內(nèi)段組 成。BDCA-2與跨膜區(qū)的 FcRγ鏈形成 adaptor 來傳遞細胞內(nèi)信號，并誘導(dǎo)類似 B 細胞受體 (BCR)的信號轉(zhuǎn)導(dǎo)級聯(lián)反應(yīng)。 抗體介導(dǎo)的 BDCA2 結(jié)合會導(dǎo)致 SYK 招募到 FcεRIγ磷酸化的免疫受體 基激活結(jié)構(gòu)域(ITAM)。SYK 激活會導(dǎo)致 BTK 和 PLCγ2 激活，從而導(dǎo)致鈣離子動員。BDCA2 受體的結(jié)合已被證明可以抑制 TLR7 或 TLR9 誘導(dǎo)的 IFN I 和其他 pDC 衍生的促炎介質(zhì)。 | |
